Description
Background:
Cetrelimab is a humanized IgG4 mAb targeting PD-1. Cetrelimab binds PD-1 (Kd=1.72 nM, HEK293) to block the interaction of PD-1 with PD-L1 and PD-L2 (IC50s=111.7 ng/mL and 138.6 ng/mL, respectively). Cetrelimab stimulates peripheral T cells, increases IFN-, IL-2, TNF- level and inhibits tumor growth in vivo.
Intended Use:

Principle:
The method employs the quantitative sandwich enzyme immunoassay technique. Antibodies to Cetrelimab are pre-coated onto microwells. Samples and standards are pipetted into microwells and human Cetrelimab present in the sample are bound by the capture antibody. Then, a HRP (horseradish peroxidase) conjugated anti-Cetrelimab antibody is pipetted and incubated. After washing microwells in order to remove any non-specific binding, the ready to use substrate solution (TMB) is added to microwells and color develops proportionally to the amount of Cetrelimab in the sample. Color development is then stopped by addition of stop solution. Absorbance is measured at 450 nm.
