Description

Background:
Complement Hemolysis 50 (CH50) assay serves as a cornerstone in the assessment of complement system function, providing valuable insights into immune response efficiency and susceptibility to infectious diseases. By measuring the serum’s ability to lyse erythrocytes, the CH50 assay evaluates the overall activity of the classical pathway of complement activation. The Human CH50 ELISA Kit offers a reliable method for quantifying CH50 levels in human serum samples, facilitating studies aimed at understanding immune system function, autoimmune disorders, and infectious diseases.

Pathways and Relevance:
The Human CH50 ELISA Kit is instrumental in elucidating pathways associated with complement activation, immune complex-mediated diseases, and host defense mechanisms. By quantifying CH50 levels, researchers can assess complement system activity, monitor disease progression, and evaluate therapeutic responses in conditions such as systemic lupus erythematosus (SLE), rheumatoid arthritis, and sepsis. Furthermore, CH50 serves as a prognostic marker for disease severity and treatment outcomes, highlighting its relevance in patient management and personalized medicine approaches.

Current Research and Ongoing Discoveries:

Researchers worldwide are leveraging the Human CH50 ELISA Kit to advance our understanding of immune regulation and complement-mediated diseases. Ongoing research endeavors include investigating the mechanisms underlying complement activation, identifying novel biomarkers for disease diagnosis and prognosis, and developing targeted therapies for complement-related disorders. Additionally, researchers are exploring the potential of CH50 as a therapeutic target in autoimmune diseases, inflammatory conditions, and infectious diseases, driving innovation and shaping the future of immunology and clinical medicine.

Validated against seven points for a “GOLD RING” Standard Quality ELISA – the benchmark sign for Krishgen Quality. The The GENLISA ELISA kits are used for assessing the specific biomarker in samples analytes which may be human serum, plasma, biological fluids and cell culture supernatant. The kit uses indirect sandwich assay with double antibodies – capture and detection to ensure a high degree of sensitivity and specificity in the estimation of Chemokine C-X-C-Motif Receptor 3 (CXCR3);

Sample Type

Serum, Plasma, Cell Culture Supernatant and other biological samples. Validated for citrated/EDTA Plasma only.

Assay Range

25 – 400 pg/ml

Sensitivity

22 pg/ml

Interference

Preparations of the known factors were assayed for interference. No significant interference was observed.

Specificity

This assay recognises both the recombinant and natural protein for 50% Complement Hemolysis (CH50)

Principle Of Assay

This ELISA is a sandwich immunoassay . Antibodies are coated on 96 well plates. The antigen protein present in sample and standard respectively bind to the coated wells. The wells are washed and an antibody:HRP Conjugate is added which binds to the bound complex in the well. Washing is performed to remove any unbound material. TMB substrate is added and the enzyme reaction is stopped by dispensing of stop solution into the wells. The optical density (OD) of the solution at 450 nm is directly proportional to the amount of antigen protein present in the standard or samples.

Detection Method

Colorimetric, Absorbance measured at 450nm

Conjugate-Enzyme Reaction

Uses stable HRP Enzyme Conjugate with (single component) TMB Chromogenic Substrate for color development.
