Multiplex ELISA technology is a natural evolution of the sandwich ELISA format — the same proven immunoassay chemistry, engineered to quantify multiple protein targets simultaneously from a single sample well. Where a standard ELISA measures one analyte per reaction, a multiplex ELISA captures 2 to 12 targets in parallel, using the same antibody-antigen-detection principle you already rely on.

The PrimePlex™ range from Krishgen BioSystems is a catalogue of fully pre-built, ready-to-use multiplex ELISA panels covering cytokines, chemokines, cardiac biomarkers, neurodegeneration markers, bone metabolism proteins, immune checkpoint molecules, and more — across 8 species including Human, Mouse, Rat, Rabbit, Guinea Pig, and Avian. With 243 pre-configured panels ranging from 2-plex to 12-plex, PrimePlex eliminates the time and technical burden of building multiplex assays from scratch.

PrimePlex kits are ideal for researchers working with limited biospecimens — clinical samples, CSF, synovial fluid, or small-volume animal bleeds — where sequential single-plex ELISA would exhaust the sample before the experiment is complete. Every panel is lot-validated and ships with a Certificate of Analysis confirming sensitivity, dynamic range, and inter-assay precision.

ELISA CHEMISTRY YOU TRUST

PrimePlex kits are built on the same sandwich ELISA immunoassay chemistry that underpins the Krishgen KRIBIOLISA range. No new platform to learn, no new reader to purchase. If your lab runs ELISA today, you can run PrimePlex tomorrow — with more data from less sample.

Performance Characteristics and Validation

Sensitivity
Each analyte channel is validated to its lowest detectable concentration — defined as the mean signal of zero standard plus 2 standard deviations. Detection limits are analyte-specific and stated in the individual kit datasheet and CoA.

Precision
Intra- and inter-assay coefficients of variation (CVs) are determined by testing high, mid, and low concentration samples across multiple runs and operators. Target CVs are <15% for all analyte channels. Lot-specific CV data are included in the Certificate of Analysis.

Specificity
Each capture-detection antibody pair is validated against structurally related family members and endogenous matrix proteins to confirm analyte selectivity in the relevant species matrix. Cross-reactivity data are provided in the datasheet.

Sample Compatibility
PrimePlex kits are optimised for serum and EDTA, heparin, or citrate plasma. Compatibility with cell culture supernatants, tissue lysates, CSF, urine, and other matrices is noted per panel. Sample-specific dilution recommendations are provided in each protocol.

Frequently Asked Questions

What is a multiplex ELISA kit and how does it differ from a standard ELISA?

A multiplex ELISA kit quantifies multiple protein targets simultaneously from a single sample well, using the same sandwich immunoassay principle as a standard ELISA. Where a conventional ELISA measures one analyte per reaction, a multiplex ELISA uses sets of matched capture and detection antibody pairs — one per target — configured to work without cross-reactivity in the same well. The result is a full multi-analyte dataset from a fraction of the sample volume that sequential single-plex assays would require.

Do I need special equipment to run PrimePlex multiplex ELISA kits?

No. PrimePlex kits are designed to run on standard 96-well ELISA plate readers — the same equipment used for conventional ELISAs. No luminex instrument, flow cytometer, or specialist multiplexing platform is required. If your laboratory currently runs ELISA, you have everything you need to run PrimePlex panels from day one.

Which species are PrimePlex multiplex ELISA kits available for?

PrimePlex panels are currently available for Human, Mouse, Rat, Rabbit, Guinea Pig, Avian (Gallus gallus), Sheep, and a General cross-species format for select endocrine analytes. The species breadth makes PrimePlex suitable for both translational clinical research and a wide range of preclinical model systems.

How many analytes can I measure in a single PrimePlex kit?

PrimePlex panels range from 2-plex (two simultaneous analytes) to 12-plex (twelve analytes in one well). The panel size is selected based on the biological pathway being profiled — smaller panels for focused mechanistic studies, larger panels for broad discovery or pathway mapping. All pre-built configurations are validated as complete units; analyte channels are not sold separately.

Can I use PrimePlex kits with cell culture supernatants or tissue lysates?

PrimePlex panels range from 2-plex (two simultaneous analytes) to 12-plex (twelve analytes in one well). The panel size is selected based on the biological pathway being profiled — smaller panels for focused mechanistic studies, larger panels for broad discovery or pathway mapping. All pre-built configurations are validated as complete units; analyte channels are not sold separately.

Can I use PrimePlex kits with cell culture supernatants or tissue lysates?

Yes. While PrimePlex panels are primarily optimised and validated for serum and plasma (EDTA, heparin, citrate), many panels are also compatible with cell culture supernatants, tissue homogenates, BAL fluid, CSF, urine, and other biological matrices. Specific matrix compatibility and recommended dilution factors are noted in each kit's datasheet and protocol.

Can Krishgen configure a custom multiplex ELISA panel for my specific targets?

Yes. If your required combination of targets is not available in the standard 243-panel catalogue, Krishgen's technical team can discuss custom multiplex panel configurations.
