Background: Cluster of differentiation 86 (CD86), also known as B-lymphocyte activation antigen B7-2, is a type I membrane protein that belongs to the immunoglobulin superfamily and is constitutively expressed on interdigital dendritic cells, Langerhans cells, peripheral blood dendritic cells. cells, memory B cells and germinal center B cells. In addition, B72 is expressed at low levels in monocytes and can be regulated by interferon γ. CD86 is a ligand for two different proteins on the surface of the T-cell: CD28 (for autoregulation and cell-cell association) and CTLA-4 (for regulation and inhibition of cell detachment). CD86 works with CD80 to prime T cells. A recent study showed that B7-2 promotes the formation of a mature APC pool and promotes APC function and survival. In addition, B7 proteins are also involved in innate immune responses by activating the NF-κB signaling pathway in macrophages. CD86 is thus considered a promising candidate for immunotherapy. CD86+ macrophages in patients with Hodgkin’s lymphoma is an independent marker of possible non-response to first-line therapy. The KRIBIOLISA Inhibitor Assay Kits available enable researchers to test the inhibitory effects of various compounds on specific enzymes at a higher output. These assays can help identify or characterize enzyme inhibitors, which is important in early drug discovery and development. They can also be useful in characterizing enzyme activity. These Inhibitor assay kits contain a known inhibitor as a control.

This Assay kit is designed to facilitate the identification and characterization of B7-2 pathway inhibitors.

Assay Features include:
– Ready to use protocol with pre-coated breakapart wells for ease of use
– Standardisation and High Reproducibility
– Lot to Lot Consistency
– Accuracy and Precision

Validated against seven points for a Gold Ring Standard Quality – the benchmark sign for Krishgen quality.
The kits are used for assessing the specific biomarker in samples analyte which may be serum, plasma, biological fluids and cell culture supernatant.
Unlike competitor assays available, we offer pre-coated ready-to-use assays to ensure high level of reproducibility and accuracy.

Principle Of Assay

The KRIBIOLISA Inhibitor ELISA is based on the Competitive/inhibition principle. A target antigen or receptor is immobilized on the microwells.. Test inhibitors compete with a labeled detection agent (e.g., enzyme-linked antibody or antigen) for binding sites, lowering the colorimetric signal proportionally to inhibitor potency. This interference-based readout distinguishes inhibitors from non-inhibitors.

Colorimetric, Absorbance measured at 450nm

Uses HRP enzyme conjugate with TMB chromogenic substrate for color development

Interference

Preparations of the known factors were assayed for interference. No significant interference was observed.

Specificity

This assay recognises both the recombinant and natural protein
