Background: Herpesvirus entry mediator (HVEM), also known as TNFRSF14, TR2 (TNF receptor-like molecule) and ATAR (another TRAF-related receptor), is a type I transmembrane protein member of the TNF receptor superfamily. It is expressed in many immune cells, including T and B cells, NK cells, monocytes and neutrophils. Two TNF superfamily ligands, lymphotoxin alpha (TNF-beta) and LIGHT (TNFSF14), are identified as cellular ligands for HVEM and initiate positive signaling. However, recent studies have shown that HVEM also participates in a unique T-cell inhibitory signaling pathway by activating tyrosine phosphorylation of the immunoreceptor tyrosine-based inhibitory motif (ITIM) in the suppressor of B and T lymphocytes (BTLA). HVEM provides a stimulatory signal after T-cell engagement with LUMO (TNFSF14). Conversely, it can also provide an inhibitory signal to T cells when it binds suppressor of B and T lymphocytes (BTLA), a ligand member of the immunoglobulin (Ig) superfamily. Thus, HVEM can be considered a molecular switch capable of facilitating both stimulatory and inhibitory co-signaling in T cells.
